The unfolded protein response is the mechanism by which cells control endoplasmic reticulum (ER) homeostasis. Under normal conditions, UPR not activated; however, under certain stresses, such as hypoxia or altered glycosylation, can be activated due to an accumulation of proteins. activation involves three signaling pathways, IRE1, PERK and ATF6, all play vital roles in returning homeostasis levels seen non-stressed cells. IRE1 best studied it only pathway present Saccharomyces cerevisiae. This spliceosome independent splicing HAC1 XBP1 yeast mammalians cells, respectively. limits synthesis, therefore reducing number new proteins requiring folding. ATF6 translocated proteolytically cleaved, releasing a NH2 domain fragment transported nucleus affects gene expression. If unsuccessful at load ER signals remain activated, this lead programmed cell death.
